The 31-year-old pop star died after falling from a third-floor balcony at the Casa Sur hotel in Buenos Aires on October 16.
Those arrested are accused of " abandonment of a person followed by death, supply of narcotics" , according to a statement from the office of the prosecutor Andres Esteban Madrea.
The statement said that " Liam Payne was not fully conscious or experiencing a state of obvious collapse or loss of consciousness at the time of the fall."
One of the accused " accompanyed the artist on a daily basis " during his stay in the Argentine capital, while another was a hotel employee.
The prosecution said it had taken dozens of testimonies, analyzed more than 800 hours of video from security cameras and public streets, and analyzed the contents of Payne's phone, allowing his calls, messages and social media networks to be analyzed.
